氟虫腈单克隆抗体制备及其性能分析 被引量:4

摘要  利用氟虫腈类似物FH作为半抗原,与蛋白偶联后免疫Balb/c小鼠,取其脾脏细胞与SP2/0骨髓瘤细胞融合,得到产生阳性抗体的杂交瘤细胞株3C3。经过各项条件优化建立的间接竞争ELISA方法对氟虫腈最低检测浓度为0.074ng·ml-1,抑制中浓度为5.99ng·ml-1。抗体与定虫隆、三氟氯氰菊酯、氟乐灵、氟虫脲的交叉反应率小于0.67%。对添加到两种不同基质中的氟虫腈进行检测,结果变异度为1.8%~9.8%,回收率为94.7%~108.0%。 One hybridomacell lines 3C3 secreting monoclonal antibodies against insecticide fipronil was prepared by using mergenced myeloma cells SP2/0 of mouse with spleen cells from Balb/c mice immunized by the hapten fipronil analogue conjugated bovine serum albumin.The effect of several physicochemical factors on the monoclonal antibody for fipronil was studied and the results were used to optimize the assay system. The most low limited quality was 0.074 ng·ml-1,and the IC50 was 5.99 ng·ml-1 measured by an indirect competitive ELISA.Crossreactivities on Chlorfluazuron,Cyhalothrin,Trifluralin and Flufenoxuron were less than 0.67%.The range of recovery rates of fipronil from two matrixes was 94.7%~1080% and the variation coefficient was 1.8%~9.8%.So monitoring fipronil by using monoclonal antibody is capable.
