运用原位杂交和二步法技术检测乳腺癌PTTG基因表达 被引量:2

Expression of PTTG(Pituitary Tumor Transforming Gene) in Human Breast Carcinoma

摘要 目的 研究PTTG基因在乳腺癌组织中表达的临床意义。方法 应用原位杂交和二步法 ,检测 66例乳腺癌、2 7例乳腺小叶增生及 5 4例乳腺正常组织中PTTGmRNA和PTTG蛋白的表达。结果 PTTGmRNA在乳腺癌、乳腺小叶增生和乳腺正常组织中表达阳性率分别为 71.2 % (4 7/66)、5 1.9% (14 /2 7)和 11.1% (6/5 4) ,而PTTG蛋白表达阳性率分别为 63 .6% (4 2 /66)、48.1% (13 /2 7)和 7.4% (4 /5 4)。乳腺癌、乳腺小叶增生与乳腺正常组织之间的PTTGmRNA及PTTG蛋白阳性率比较均有显著性差异 (P <0 .0 5 ) ;PTTGmRNA的表达与PTTG蛋白表达呈正相关 (P <0 .0 5 ) ,而与淋巴结转移无关 (P >0 .0 5 )。 Objective To investigate the significance of PTTG expression in human breast carcinomas(BRCA).Methods In situ hybridization(DNA-RNA) and PV-9000 polymer detection system were used to detect the expression of PTTG mRNA and protein in 66 cases of BRCA,27 cases of breast lobule hyperplasia and 54 normal controls.Results The positive rates of PTTG mRNA and protein were 71.2%(47/66) and 63.6%(42/66) in BRCA,51.9%(14/27) and 48.1%(13/27) in breast lobule hyperplasia tissue,11.1%(6/54)and 7.4%(4/54)in breast normal tissue respectively.The expression rate was significantly higher in BRCA and breast lobule hyperplasia tissue than in breast normal tissue(P<0.05).The expression of PTTG mRNA was significantly correlated with that of PTTG protein(P<0.05),but no significant correlation was observed between the expression of PTTG and the metastasis of lymph node(P>0.05).Conclusion Over-expression of oncogene PTTG may be related to human breast carcinogenesis.
