摘要
报道了利用大肠杆菌氨基酰化酶Ⅰ拆分消旋体生产L氨基酸的研究结果。大肠杆菌氨基酰化酶Ⅰ ,在培养 2 0~ 2 2小时 ,酶活力可达到 89.2 μmol/ml·h- 1。用它拆分DL Met ,其最适反应温度为 6 0℃ ,最适 pH值为 7.0 ,最适底物浓度为 0 .2 5~ 0 .2 75mol/L ,催化动力学过程不符合米式方程。 0 .0 4mol /L的Co2 + 对酶有较好的激活作用 ,活力提高
There is carbamylation enzyme I in E.coli. After E.coli has been cultiveted for 20~22 hours, the enzyme activity will come to 89.2 umol/mlh -1 .Being used to disperse DL Met,the optimum temperature is 60℃,optimum pH is 7.0 and optimum substrate conceration is 0.25~0.275mol/L.And it's catalysis dynamics is not apt to Michaelis reaction.At the concentration of 0.04 mol/L, Co 2+ has a good activation so that enzyme activity is lifted by 8% or so.
