摘要
建立了用高效液相色谱法对氢化可的松发酵液中的主要组分氢化可的松(HC)、表氢化可的松(EHC)、17α 羟基孕甾 4 烯 3,20 二酮 21 醋酸酯(RSA)和17α 羟基孕甾 4 烯 3,20 二酮(RS)进行测定的方法。采用硅胶吸附色谱柱,以二氯甲烷 乙醚 甲醇 水(体积比为385∶60∶30∶2)为流动相,流速0 8mL/min,在242nm波长下检测。发酵液各组分分离良好,HC,EHC,RSA和RS的线性范围分别为25~375mg/L,5~75mg/L,5~75mg/L和5~75mg/L;检出限分别为0 4,0 1,0 05和0 02mg/L。方法准确,灵敏,重现性好。
A method for the determination of hydrocortisone (HC), epihydrocortisone (EHC), 17α-hydroxypregn-4-en-3,20-dione-21-acetate (RSA) and 17α -hydroxypregn-4-en-3,20-dione (RS) in the culture by high performance l iquid chromatography (HPLC) was developed. The HPLC was performed on a Phenomene x Prodigy Silica 100A column (5 μm, 250 mm×46 mm id) by using dichlorome thane-ether-methanol-water (385∶60∶30∶2, v/v) as mobile phase. The flow r ate was 08 mL/min and the UV detection wavelenth was 242 nm. The method ha s been successfully used for the determination of the four compounds with rec overies of 1009%, 968%, 1005% and 1006% for HC, EHC, RSA and RS, respect ively. The linear ranges of HC, EHC, RSA and RS were 25-375 mg/L, 5-75 mg/ L,5-75 mg/L and 5-75 mg/L with the detection limits of 04, 01, 0 05 and 002 mg/L, respectively. This method is accurate, sensitive and reproducible.
