胰岛素与福辛普利对自发性高血压大鼠心肌胰岛素样生长因子-1受体基因转录的影响 被引量:1

摘要 目的 :观察长期注射中效胰岛素和喂食血管紧张素转换酶抑制剂福辛普利对自发性高血压大鼠 (SHR)血压、血浆胰岛素浓度及心肌胰岛素样生长因子 1受体基因转录的影响。  方法 :3 2只 10周龄SHR随机分 4组 :1组 :注射中效胰岛素 5U/(kg·d) ,服福辛普利 5mg/(kg·d) ;2组 :注射蒸馏水 0 0 5ml/(kg·d) ,服福辛普利 5mg/(kg·d) ;3组 :注射中效胰岛素 5U/(kg·d) ,服安慰剂 5mg/(g·d) ;4组 :注射蒸馏水 0 0 5ml/(kg·d) ,服安慰剂 5mg/(kg·d)。实验前及实验中每 2周测鼠尾动脉血压 ,9周后抽血并处死动物取材 ,测量血浆胰岛素浓度和心肌胰岛素样生长因子 1受体的信使核糖核酸水平。  结果 :SHR血浆胰岛素浓度注射中效胰岛素较未注射的高 ,有极显著性差异 (P <0 0 1) ;SHR的血压、心肌胰岛素样生长因子 1受体的信使核糖核酸水平 ,注射胰岛素与否 ,均没有显著性差异 (P >0 0 5 ) ;SHR血压、心肌胰岛素样生长因子 1受体的信使核糖核酸水平用福辛普利较用安慰剂SHR的低 ,均有显著性差异 (P <0 0 5 )。  结论 :长期注射中效胰岛素可引起SHR高胰岛素血症 ,但不影响其血压及心肌胰岛素样生长因子 1受体基因转录水平 ;而喂食福辛普利可使其胰岛素样生长因子 1受体基因转录水平下调。 Objective:To observe the influence of long-term injection of Insulin and fosinopril feeding on blood pressure and insulin-like growth factor-1 receptor gene transcription in spontaneously hypertensive rats(SHR). Methods:Thirty-two ten-week-old SHR were divided randomly into 4 groups,group 1:injection of insulin [5 U/(kg·d)] and fosinopril feeding [5 mg/(kg·d)],group 2:injection of distilled water [0.05 ml/(kg·d)] and fosinopril feeding [5 mg/(kg·d)],Group 3:injection of insulin [5 U/(kg·d)] and placebo feeding [5 mg/(kg·d)],group 4:injection of distilled water [0.05 ml/(kg·d)] and placebo feeding [5 mg/(kg·d)].Before and after experiment,BP was measured by caudal artery method every 2 weeks.After 9 weeks the blood and total mRNA were extracted from myocardium of SHR.The level of plasma insulin and myocardium insulin-like growth factor-1 receptor mRNA were measured. Results:Plasma insulin level of insulin injected groups was significantly higher than those of non-injected groups (p<0.01).There were no differences in BP and insulin-like growth factor-1 receptor mRNA level of myocardium between injected insulin group and non-injected group(p>0.05).The BP and insulin-like growth factor-1 receptor mRNA level of fosinopril fed groups were significantly lower than those of placebo group (p<0.05). Conclusion:Long-term injection of insulin can increase insulin level but not influence BP and gene transcription of myocardium insulin-like growth factor-1 receptor in SHR. BP and gene transcription of myocardium insulin-like growth factor-1 receptor in SHR were declined by fosinopril feeding.
