庆大霉素中毒性急性肾小管坏死大鼠模型的建立及观察 被引量:9

摘要 给成年Wistar大鼠皮下注射庆大霉素(GM)400 mg.kg-1.d-1×2 d建立GM中毒性急性肾小管坏死(ATN)动物模型,同时观察重组人表皮生长因子(rhEGF)对其肾功能和组织形态的影响。注射GM后第5天近端小管出现广泛坏死,随后逐渐恢复。EGF治疗组肾功能较GM组提前7天恢复至正常水平;肾损伤早期两组病理形态差别不明显,EGF治疗组较GM组提前7天恢复正常组织形态。结果提示,rhEGF具有促进GM中毒性ATN形态和功能恢复的作用,但无保护作用;其促进恢复的作用与肾小管损伤程度及所用剂量有关。 Acute tubular necrosis animal models were established by subcutaneous injection of gentamicin(GM) 400 mg/kg/day for 2 days in adult Wistar rats and the effects of recombinant human epidermal growth factor (rhEGF) on the renal function and histology were observed. On 5th day after the first GM exposure, epithelial cells of proximal tubules were diffusely necrotic and then gradually recovered to normal on 21 th day. Renal function values of EGF-treated animals returned to normal 7 days earlier than that of GM-treated animals. At the early stage of renal injury, the histological changes were not different from each other. When examined at days 11 and 15, kidneys from rats that received rhEGF were improved in histopahtological appearances compared with those from GM-treated animals. These results suggests that the rhEGF enhances renal tubular repair and accelerates the recovery of renal function in a model of gentamicin nepro-toxicity in rats, and the effects of rhEGF are related with a certain degree of injury of the models and its dosage.
