肌肉注射灭活SARS病毒早期对BALB/c小鼠黏膜论著部位淋巴细胞表型的影响

Effect of inactivated SARS virus on the lymphocyte phenotypes from different mucosal tissues in BALB/c mice by intramuscular injection during the early stage

摘要 目的 :了解灭活SARS病毒肌肉途径注射小鼠后早期对小鼠黏膜免疫系统的影响。方法 :将 5周龄的雌性BALB/c小鼠随机分为两组 ,每组 5只 ,实验组每只小鼠经肌肉注射溶于 1 0 0 μlPBS中的 5 0 μg纯化的灭活SARS病毒 ,对照组注射等量的PBS ,3d后处死小鼠 ,用流式细胞仪测定不同黏膜组织中淋巴细胞表型的变化。结果 :肌肉注射灭活SARS病毒使鼻黏膜相关淋巴组织 (NALT)中CD+ 4 荧光强度显著增加 (P <0 .0 5 )、CD+ 3 荧光强度有增加趋势 (P =0 .0 5 2 ) ;鼻通道 (NP)淋巴细胞中CD+ 4 和CD+ 8构成比显著增加 (P <0 .0 5 )、CD+ 3 构成比和T/B比值有增加趋势 ,而CD+ 4 ,CD+ 3 和B细胞荧光强度都有显著增加 (P <0 .0 5 ) ;肺CD+ 8构成比显著降低 (P <0 .0 5 )、B细胞构成比显著增加 (P <0 .0 1 )、CD+ 4 构成比有降低趋势 (P =0 .2 38)、T/B比值有降低趋势 (P =0 .0 77)、CD+ 4 /CD+ 8比值有增加趋势 ,而CD+ 3 荧光强度显著增加 (P <0 .0 5 )、CD+ 4 荧光强度有增加趋势 (P =0 .0 86 ) ;小肠派氏集合淋巴结 (PP)中B细胞构成比显著增加 (P <0 .0 5 )。结论 :肌肉注射SARS灭活病毒早期 ,可以使多部位黏膜淋巴组织的淋巴细胞表型发生变化 。 Objective:To observe the effects of inactivated SARS virus on the mucosal immune system in BALB/c mice by intramuscular injection during the early stage in mice. Methods:Five week_old female BALB/c mice were randomly divided into two groups, five mice per group. Each mouse of the experiment group was intramuscularly injected with a single dose of 50?μg of the inactivated SARS virus in 100?μl PBS without any adjuvant, the control ones were injected into the equal volume of PBS. Then all mice were sacrificed on the third day, and the lymphocyte phenotypes from different mucosal tissues were detected by FACS.Results:After intramuscular injection with inactivated SARS virus, the fluorescent intensity of CD + 4 T cell in nasal mucosa associated lymphatic tissue (NALT) strengthen significantly( P < 0.05) on the third day, and that of CD + 3 T cell in NALT tended to increase ( P =0.052). The CD + 4 and CD + 8 T cells in nasal passage (NP) increased significantly( P <0.05) than that of control ones. Both the CD + 3T and the ratio of T/ B in NP had an increased trend, whereas the fluorescent intensity of CD + 4,CD + 3T and B cells in NP strengthen significantly than that of the control mice ( P <0.05). In the lung, CD + 8T cells decreased significantly( P < 0.05) , but B cells increased significantly ( P <0.01). The CD + 4 T and the ratio of T/ B from lung had a decreased trend, whereas the ratio of CD + 4/CD + 8 tended to increase. Not only the fluorescent intensities of CD + 3T cells in lung enhanced ( P <0.05), but also the CD + 4T cells in lung had an increased trend ( P =0.086). On the other hand, B cells in Peyer′s patches(PP) increased significantly ( P <0.05) than that of control mice.Conclusions:The lymphocyte phenotype from different mucosal tissues of BALB/c mice changed after intramuscular injection with inactivated SARS virus on day 3, suggesting that the inactivated SARS virus could induce mucosal immune response in the intramuscularly injected mice.
