摘要
目的 :研究苦参碱 (matrine)对血管紧张素Ⅱ (AngⅡ )诱导新生大鼠心肌成纤维细胞 (cardialfibroblasts,CFb)增殖和胶原合成的影响 ,探讨其抑制心肌纤维化的机制。方法 :建立AngⅡ诱导新生大鼠CFb纤维化模型 ,采用MTT法检测Mat对CFb增殖的影响 ;羟脯氨酸测定检测CFB胶原合成量 ;RT PCR检测Mat作用下CFb的Ⅰ型胶原、间质胶原酶 (MMP 13)、组织金属白酶抑制因子 1(TIMP 1)、TGFβ 1的基因表达情况。结果 :①在一定范围内 ,苦参碱以浓度依赖性方式抑制AngⅡ诱导CFb的增殖和胶原合成 (P <0 .0 1) ,②苦参碱可以降低Ⅰ型胶原和TGFβ 1基因mRNA表达 ,③苦参碱可以提高MMP 13基因mRNA表达。结论 :苦参碱通过抑制AngⅡ诱导的CFb增殖和胶原合成增加 。
Objective: To study the influence of matrine on the preliferation and collagen metabolism of cultured neonatal ratcardic fibroblast (CFb) stimulated by angiotensin Ⅱ,and to explore the mechanism of matrine on cardic fibrosis. Methods: Neonatal rat cardic fibroblast were treated with angiotensin Ⅱ(AngⅡ) to form fibrosis model. The effects of matrine on proliferation of CFb were observed by MTT colorimetric assay,synthesis of collagen was observed by the hydroxyproline concentration measured. Collagen Ⅰ, matrix metalloproteinase(MMP 13), the tissue inhibitor metalloproteinase (Timp 1) and TGFβ 1 mRNA expression were examined by semi quantitatative RT PCR analysis. Results: ①Within a concentration coverage,matrine (1.00~0.125 mmol·L -1 ) inhibited CFb proliferation and collagen synthsis stimulated by angiotensin Ⅱby a dose dependent way (P<0.01). ②Matrine decreased the levels of Collagen Ⅰand TGFβ 1 mRNA expression. ③ Matrine increased the levels of MMP 13 mRNA expression. Conclusion: Matrine inhibited cardic fibrosis stimulated by angiotensin Ⅱby inhibiting CFb proliferation and collagen synthesis.
