化合物N-2035对大鼠局灶性脑缺血再灌注损伤的保护作用 被引量:7

摘要 目的:观察异黄酮类化合物N-2035对大鼠局灶性脑缺血再灌注损伤的保护作用。方法:利用大脑中动脉栓线阻断法(MCAO)制备大鼠局灶性脑缺血再灌注损伤模型,观察化合物N-2035对缺血再灌注大鼠的脑水肿、细胞内钙及凋亡抑制基因bcl-2表达的影响。结果:化合物N-2035能改善局灶性脑缺血再灌注损伤脑水肿,降低细胞内钙,增加bcl-2的表达。结论:化合物N-2035对大鼠局灶性脑缺血再灌注损伤具有保护作用,其作用机制可能与化合物N-2035抗细胞凋亡有关。 Objective:To investigate the protective effect of compound N-2035obtained from puerarin,a kind of isoflavone that was extracted from Chinese traditional medicine on focal brain is-chemia-reperfusion injury in rats.Methods:The models of focal brain ischemia reperfusion injury by middle cerebral artery occlusion(MCAO)were established in Wister rats.Cerebral edema was measured by dry /wet mass method and intracellular calcium and the expression of bcl-2was examined by flow cytometer.Re sults:N-2035in a dose of40mg /kg positively relieved cerebral edema;each dose de-creased intracellular calcium and increased the expression of bcl-2.Con clusion:Compound N-2035can relieve brain damage induced by focal ischemia /reperfusion in rats.The mechanism may be related with the decrease of intracellular calcium and the increase of the expression of bcl-2.
