摘要
目的 观察组蛋白去乙酰化酶抑制剂三丁酸甘油酯 (tributyrin ,TB)对K5 6 2白血病细胞增殖、凋亡的影响 ,并进一步探讨其作用机制。方法 用细胞计数及台盼兰拒染法观察TB对K5 6 2细胞增殖及活力的影响 ,通过细胞形态观察、流式细胞术分析观察TB对K5 6 2细胞体外诱导凋亡的情况 ,RT -PCR技术分析 p2 1WAF1表达的改变。结果 (1)TB抑制K5 6 2细胞的增殖。 (2 )TB诱导K5 6 2白血病细胞的凋亡 ,影响细胞周期的进程 ,使细胞周期阻滞于G2 /M期。 (3)在TB引起的K5 6 2细胞增殖抑制及凋亡过程中 ,p2 1WAF1表达增加。结论 TB抑制K5 6 2白血病细胞的增殖并诱导凋亡 ,阻滞细胞周期进程于G2 /M期 ,p2 1WAF1在此过程中发挥作用。
Objective To investigate the effect of tributyrin (TB), a histone deacetylase inhibitor (HDACi) on the cell growth and apoptosis in K562 cell and elucidate its mechanism. Methods Cell proliferation and viability were analyzed by cell count and Trypan blue exclusion assay. Cell morpholo- gical analysis, flow cytometry and semi-quantitative RT-PCR were performed to determine whether TB could induce apoptosis of K562 cell in vitro and the change of p21 WAF1 expression. Results (1) TB inhibited K562 cell growth. (2) TB induced apoptosis in K562 cell and affected the cell cycle progression, most of cells were arrested in G2/M phase. (3) p21 WAF1 mRNA were up regulated by TB. Conclusion TB can inhibit proliferation, induce apoptosis, and interfere with cell cycle progression in K562 cell. p21 WAF1 may be involved in the course of the TB-induced apoptosis.
