摘要
目的研究TRAIL对胃癌细胞株MKN28细胞体外诱导凋亡的作用及机制,并研究此过程中Caspase-3的表达规律及与凋亡效应的相关性。方法培养细胞,MTT法分析细胞增殖抑制率及细胞存活率,流式细胞仪分析细胞凋亡及细胞周期,细胞免疫组化检测细胞Caspase-3的表达水平,AO/EB染色观察凋亡细胞。结果 TRAIL可明显抑制MKN28细胞的增殖,并可诱导细胞的凋亡。细胞阻滞于G0/G1期,Caspase-3在MKN28细胞中低表达,经TRAIL处理细胞后高表达。AO/EB染色后可见典型细胞凋亡特征。结论 TRAIL可诱导MKN28细胞株的凋亡,并呈剂量依赖性。TRAIL通过上调Caspase-3的表达,阻滞细胞周期,诱导MKN28细胞的凋亡。
Objective:To investigate apoptosis induction of TRAIL and its mechanism in gastric cancer cell MKN28 line in vitro,and to study the express of Caspase-3 and its correlation with apoptosis.Methods: Their influence on cell proliferative activity was detected by MTT assay.The cell apoptosis and cell cycle was determined by flow cytometric(FCM) analysis.The expression of Caspase-3 was detected by immunohistochemistry.apoptosis was detected by AO/EB staining.Results: TRAIL could obviously inhibit the proliferation of MKN28 cells,and induce its apoptosis.Cell cycle was arrested in the G0/G1 phase.After treatment of TRAIL,expression of Caspase-3 was induced in MKN28 cells.MKN28 cells showed the typical features of apoptosis after AO/EB staining.Conclusion: TRAIL had effects of inducing apoptosis on MKN28 cell lines with a dose-dependence.The apoptosis of MKN28 cells was induced by TRAIL through inducing expression of Caspase-3 and arresting cell cycle.
