药代动力学/药效学参数:优化抗菌药物给药方案 被引量:23

摘要 临床应用抗菌药物治疗的目的是清除病原菌,以获得最大疗效并使不良反应降至最低,此外要减少耐药菌的产生.为了达到这个目的,需要正确理解和应用药代动力学(PK)和药效学(PD)原理,设计最佳给药方案.传统上用体外获得的静态数据如最低抑菌浓度(MIC)和最低杀菌浓度(MBC)指导抗菌治疗.对临床医生来说,MIC仅仅是病原菌对药物敏感性的一个指标,忽略了抗菌药物在体内按一定规律变化的特点.越来越多研究表明MIC与临床结果不完全一致,不能很好预测临床结果,因此,根据MIC设计给药方案并不是理想方法[1-3].
