摘要
目的 研究高压氧 (HBO)治疗对急性 CO中毒大鼠海马区神经元 Bc1- 2蛋白表达的影响。方法 6 0只雄性 SD大鼠随机分成 3个组 (正常对照组、CO中毒不治疗组、CO中毒 HBO治疗组 )。各组大鼠分别暴露在空气或 CO气体 (3.2× 10 - 3体积分数 )中 6 0 m in,对 CO中毒 HBO治疗组大鼠行HBO治疗 ,常规 HE染色和免疫组化染色 ,观察 CO中毒后第 1,3,5和 7天大鼠海马区神经元损伤和Bc1- 2蛋白表达变化的特点。结果 CO中毒组大鼠海马可见大量变性坏死神经元 ,HBO治疗组海马区神经元变性坏死减少 ,Bc1- 2蛋白表达增多 ,尤以中毒后 3和 5 d明显 (P<0 .0 5 )。结论 HBO治疗可以促进急性 CO中毒大鼠海马区神经元 Bc1- 2蛋白表达 ,具有保护神经元的作用。
Objective To investigate the effect of hyperbaric oxygen (HBO) treatment on the expression of protein Bc1-2 in the rat hippocampus after acute carbon monoxide poisoning.Methods 60 Sprague-Dawley male rats were randomly divided into three groups (control group,CO-exposed group and HBO-treated group).The control group,CO-exposed group and HBO-treated group were exposed to either air or CO(3.2×10 -3 volume fraction) for 60 min.Rats of the HBO-treated group were treated by 0.25 MPa HBO.General hematoxylin-eosin(HE) stain and immunohistochemistry stain were used to observe the neurological damage and the change of Bc1-2 expression in the rat hippocampus on day 1,3,5 and 7 after CO exposure.Results In the hippocampus of HBO-treated group,the number of necrotic neurons was significantly decreased and the expression of protein Bc1-2 was increased,compared with the CO-exposed group.Especially,on day 3 and 5 after CO-exposure,the expression of protein Bc1-2 was significantly increased (P<0.05).Conclusions HBO treatment can promote the expression of protein Bc1-2 in the rat hippocampus following acute carbon monoxide poisoning and protect neuronal cells.
