摘要
目的观察细胞凋亡因子Fas抗原与阿尔茨海默病(Alzheimerdisease,AD)脑神经细胞凋亡的关系。方法采用免疫组织化学的方法,观察Fas抗原及β淀粉样蛋白(β-AP)在6例AD,5例非痴呆老年人(ND)大脑皮层、海马、嗅球等部位的表达。结果在AD脑内Fas抗原主要表达于大脑皮质区的神经元胞膜上,而在ND脑内仅有少量或没有神经细胞呈Fas抗原阳性(P<0.01)。同时,我们发现脑皮层Fas抗原与β-AP的分布及阳性度之间呈紧密相关。结论AD脑内神经细胞的丢失可能与Fas抗原介导的细胞凋亡机制相关,并且与β-AP的沉积有关。
Objective To reveal the correlations between cell apoptosis factor Fas antigen and neuron
apoptosis in the brains of the patients with Alzheimers disease (AD). Methods Using
immunohistochemistry (IHC) technology, we obsered the expression of Fas antigen and β
amyloid protein (β AP) at different regions of cerebral cortex, hippocampus and olfactory ball
in 11 cases autopsy brain, including 6 cases of AD and 5 of non dementia (ND). Results In AD
brains, Fas antigen was mainly expressed in neuron membrane of new cortex. But there was
only less or no expression of Fas antigen in neurons of ND ( P <0.01). In all brains, the
amount and the distribution of Fas antigen expression were closely correlated to those of
expression of β AP.Conclusions The pathogenesis of Fas mediated apoptosis may be
association with neuron decrease and β AP deposited in AD brain.
