摘要
目的 :通过提高肿瘤细胞活性氧 (ROS)水平 ,探讨增加三氧化二砷 (As2 O3)促进细胞凋亡的敏感性。方法 :As2 O3与天然醌类物质大黄素和醌类氧化酶抑制剂双香豆素合用 ,作用于 He L a细胞 ,分别以 MTT法和流式细胞仪检测细胞活力、细胞凋亡率和活性氧水平。结果 :大黄素 10μm ol/ L通过提高 ROS增强了 He L a细胞对 As2 O3的敏感性 ,双香豆素进一步增强了这一作用。结论 :提高肿瘤细胞 ROS水平可能是提高 As2 O3疗效的一种手段。
Objective:To enhance the susceptibility of tumor cells to arsenic trioxide (As 2O 3)-induced apoptosis through elevating cellular reactive oxygen species (ROS) level. Methods:Cell proliferation assay and flow cytometry were used to assess cell viability, apoptosis and the cellular ROS level after treatment with As 2O 3 alone or in combination with natural anthraquinone emodin and quinone reductase inhibitor dicoumarol.Results:Emodin(10 μmol/L) sensitized Hela cells to As 2O 3-induced apoptosis via generation of ROS, and dicoumarol enhanced this effect.Conclusion:Elevating cellular ROS level might be a novel strategy to expand the chemotherapy spectrum of As 2O 3.
