汉防己甲素和阿苯达唑联合治疗小鼠继发性泡球蚴病的机理探讨 被引量:1

摘要 应用中药单体汉防己甲素和阿苯达唑单独及联合用药治疗小鼠继发性泡球蚴病 ,以研究二药协同治疗泡球蚴病的机理。小鼠继发性泡球蚴病用药物治疗 90天后 ,检测各小鼠泡球蚴湿重、胸腺重量、脾脏重量、抑囊率、血清透明质酸(HA)、IgE和血清IL 2的含量 ;并进行病理组织学和超微结构观察。结果显示汉防己甲素和阿苯达唑单独及联合用药均对小鼠泡球蚴有明显抑制作用 (抑制率分别为 44 .5 5 % ,63 .3 9%和 80 .82 % ) ,其中联合用药组明显优于单独用药组 (P <0 .0 5 ) ;接种泡球蚴的各组与空白组比较 ,胸腺普遍缩小而脾脏增大 ,但治疗组较对照组变化较小 (P <0 .0 5 ) ;小鼠血清HA ,IgE和IL 2含量在用药组与对照组比较有显著性差异 (P <0 .0 5 )。说明汉防己甲素能提高小鼠机体的免疫力 ,并对小鼠泡球蚴的生长有一定的抑制作用 ,尤其联合阿苯达唑治疗效果更好 ,说明两种药物具有协同治疗作用 。 This paper reported the effect of the alkaloid Tetrandrine from Chinese herbs on experimental secondary Echinococcus multilocularis in mice and synergistic effect with Albendazole and researched synergistic mechanism of Tetrandrine and Albendazole against Echinococcus multilocularis infection in mice. The experiment was devided into five groups, including Tetrandrine alone, single Albendanzole alone, combination of the two drugs, control and blank. After being treated 90 days, the mice were taken the blood from post eyeball and dissected. The major judging criteria included cyst wet weight, inhibition rate, thymus index, spleen index, serum IgE, IL 2 and Hyaloplasm acid (HA) content, histopathological changes of cyst both microscopically and electron microscopically. The results showed that the Tetrandrine alone or combination with Albendazole had obviously effect against growing of E.multilocularis cyst (Inhibition rates were 44.55% and 80.82% respectively). The thymus index of control group was lower than that of therapeutical groups, but the spleen index of its was higher than that of therapeutical groups (P<0.05). The serum IgE, serum HA content of therapeutical groups were lower than that of control group (P<0.05). But serum IL 2 was higher than that of control group (P<0.05). Tetrandrine against E.multilocularis had obvious inhibitory effect. Therefore Tetrandrine can highten immune of mice. The combination with Albendanzole had stronger inhibition action. This indicated that Tetrandrine could strengthen Albendazole's effect and had synergetic action with Albendazole against E. multilocularis.
