摘要
目的 :探讨端粒酶活性表达与细胞增殖活性和VEGF表达的关系。方法 :收集胶质瘤病例 37例 ,其中Ⅱ级组 1 5例 ,Ⅲ级组 9例 ,Ⅳ级组 1 3例。用TPAP法检测端粒酶活性表达 ,以波长 4 5 0nm/ 5 95nm读取OD值检测杂交产物。石蜡切片免疫组化Sp法标记PCNA和VEGF。Mias图象分析系统计数PCNA阳性细胞 (PCNALI)和定义肿瘤有无表达VEGF。结果 :37例胶质瘤中 1 9例 (5 1 35 % )端粒酶活性阳性表达 ,阳性率随肿瘤级别增高而增高 (P <0 0 5 ,P <0 0 5 ,P <0 0 5 )。 37例胶质瘤平均PCNALI为 5 3± 1 8% ,随肿瘤级别增高而增高 (P <0 0 5 ,P <0 0 5 ,P <0 0 5 )。端粒酶活性组的PCNALI不高于无端粒酶活性组 (P >0 0 5 )。 37例胶质瘤VEGF表达率为 86 4 9% ,VEGF表达同端粒酶活性无关 (P >0 0 5 )。结论 :胶质瘤瘤细胞端粒酶重新表达与细胞增殖事件的最终发生可能涉及更为复杂的机制 。
Objective:To evaluate the correlation of telomerase activity with cell proliferation and VEGF in patients with glioma.Methods:Thirty-seven gliomas samples were detected for telomerase activity by telomeric reprat amplification protocol(TRAP)assay and examined PCNA and VEGF by Sp immunohistochemistry staining techniques.Numbers of tumor cells positive with PCNA and VEGF were measured digitally using Mias Image Analysis System.Results:The average PCNA LI was 53±18% and the expression of VEGF was 86 49%.Statistically,the expression of telomerase activity (51 35%)was correlated with histological differentiation( P <0 05, P <0 05, P <0 05),but did not relate to PCNA LI or expression of VEGF( P >0 05, P >0 05).Conclusion:The activation of telomerase was not related to cell proliferation or genes about expression of VEGF in patients with glioma,whereas it may involve more complex mechanism.
