体外培养新生大鼠海马神经元的图象分析 被引量:2

摘要 目的:研究体外培养海马神经元不同时期的形态学变化。方法:用图象分析方法,研究体外培养的新生Wistar大鼠海马神经元在不同时间的形态、细胞数密度、面数密度、长度密度以及分支数密度的发育性变化情况。结果:体外培养的海马神经元呈梭形、锥体形、多角形多形性生长。随着培养时间的延长,细胞数密度和面数密度保持不变,突起长度密度和分支密度逐渐增加,突起上膨体样结构数密度增加。结论:体外培养的新生大鼠海马神经元在离体1~14d,随着培养时间的延长,细胞突起、分支数目以及膨体样结构逐渐增多,细胞间连接增加,呈现形态和功能成熟过程。 Objective:To study the alterations in morphology of cultured hippocampal neurons during different developing stages in vitro.Methods:The developing changes in morphology,cell num-ber density,surface number density,length density and branch number density were estimated with the image analysis system on cultured hippocampal neurons from neonatal Wistar rats.Results:The cultured hippocampal neurons showed fusiform,pyramidal and polygonal shapes.As the culture time prolonged,the length density,branch number density of the neuron and the number of enlargements borne on the pro-cess increased,while the cell volume fraction and the surface number density remained constant.Con-clusion:The branches and enlargements as well as the joints between cultured neurons increase grand-ually during in vitro day1through day14,indicating the cultured neurons undergoing a maturation in their structures and functions.
