摘要
目的 研究老年大鼠总抗氧化能力、抗氧化酶活性及一氧化氮 (NO)和白介素 (IL) 6mRNA表达的变化规律。探讨氧化应激和炎症反应在衰老过程中的作用机制。方法 选用 2月龄及 18月龄SD大鼠 ,取血清、内脏组织测定总抗氧化能力 (TAC)、超氧化物歧化酶 (SOD)、谷胱甘肽过氧化物酶 (GSH Px)和NO。取血液、脑、心、肝、肾组织提取总RNA ,用RT PCR方法测定IL 6、诱导型一氧化氮合酶 (iNOS)mRNA的表达。结果 18月龄大鼠血清及各脏器组织匀浆中TAC、SOD和GSH Px水平均显著低于 2月龄大鼠 (P <0 .0 5 ) ;NO水平则明显增高 (P <0 .0 5 ) ;IL 6和iNOSmRNA的表达也明显增高 ,并与TAC水平下降呈负相关 (P <0 .0 5 )。结论 老年大鼠机体总抗氧化能力明显降低 ,并与NO、IL 6水平增高有关 ,表明老年机体内氧化应激过高 ,介导炎症反应增强 ,造成自身脏器功能损害。提示抗氧化治疗提高TAC水平可下调氧化应激及炎症反应 。
Objective To study the changes of total antioxidant capacity, antioxidase activity and the production of inflammatory mediators and investigate the relationship between TAC IL 6 and iNOS mRNA expression in aged rats. Methods The levels of TAC, SOD, GSH Px and NO in serum, brain, heart, liver and kidney were measured by biochemical methods. The expressions of IL 6 and iNOS mRNA in blood and organs were detected by RT PCR(18 months rats as aged group and 2 months rats as control group). Results The levels of TAC, SOD, GSH Px in serum and organs were significantly lower in the aged group than those in the control group. The production of NO and the [xxk1] expressions of IL 6, iNOS mRNA were higher than those in the control group. The higher expressions of IL 6, iNOS mRNA were negatively correlated with the levels of TAC in the aged group. Conclusion The TAC level is significantly lower in the aged rats, which is correlated with the higher expression of IL 6, iNOS mRNA. It has been demonstrated that there are high oxygen stress and inflammatory response in old rats, which may cause the autodamage of the tissues. It also implies that antioxidant therapy will enhance the level of TAC and downregulate the oxygen stress and inflammatory response.
