哇巴因、地高辛与高血压发病关系的研究 被引量:4

摘要 目的 观察哇巴因、地高辛对组织钠泵α亚单位基因表达的不同影响 ,探讨哇巴因与高血压发病的可能关系。方法 给予大鼠哇巴因 30 μg·kg-1·d-1,地高辛 4 0 μg·kg-1·d-1腹腔注射 7周。用免疫组化及原位杂交方法检测心脏、肾脏、肝脏、腹主动脉和肾上腺组织钠泵α亚单位蛋白水平及mRNA水平的表达。结果 哇巴因与地高辛对上述组织尤其是动脉及肾脏的钠泵α亚单位表达的影响不同 ,哇巴因增强动脉的α2 表达 ,地高辛则抑制其表达 ;哇巴因使肾脏α1表达增强 ,地高辛则使其减弱。结论 哇巴因导致组织钠泵α亚单位基因异常表达 。 Objective To observe the changes of sodium pump αsubunit expression in rat and by comparing the respective effect of ouabain and digoxin to study the relationship between ouabain and sodium pump subunit and hypertension. Methods Low dosage of endogenous ouabain (EO) 30?μ g·kg -1 ·d -1 and digoxin (DIG) 40?μ g·kg -1 ·d -1 was administrated respectively to rats for 7 weeks. By in situ hybridization and immunohistochemical assay, sodium pump α subunit expression of major tissues of rat in protein and mRNA level were analysed. Results The effect of ouabain and DIG on sodium pump α subunit expression in tissues such as artery and kidney was different,with EO increasing α 2 expression in artery and α 1 expression in kidney in contrast to DIG decreasing its expression. Conclusion EO and DIG affect odium pump α subunit gene expression, which may play an important role in hypertensive mechanism.
