母牛分枝杆菌菌苗对哮喘豚鼠肺组织Eotaxin mRNA表达的影响 被引量:2

摘要 目的研究母牛分枝杆菌菌苗对哮喘豚鼠肺组织Eotaxin mRNA表达的影响。方法30只豚鼠随机分为生理盐水组、哮喘组及母牛分枝杆菌菌苗组,每组10只。应用卵白蛋白(OVA)建立哮喘模型,母牛分枝杆菌菌苗组每只豚鼠在OVA致敏前10 d肌注22.5μg母牛分枝杆菌菌苗。最后一次激发后,计数支气管肺泡灌洗液(BALF)中炎症细胞,观察肺组织病理学改变,应用原位杂交方法检测肺组织Eotaxin mRNA的表达。结果母牛分枝杆菌菌苗组豚鼠BALF中嗜酸粒细胞数量显著低于哮喘组(P<0.01),肺组织哮喘炎症反应明显轻于哮喘组,肺组织Eotaxin mRNA表达(OD值)显著低于哮喘组(P<0.01)。结论母牛分枝杆菌菌苗能减少哮喘豚鼠BALF及肺组织嗜酸粒细胞数量,与其抑制Eotaxin mRNA在哮喘豚鼠肺组织的表达有关。 Objective To study the effect of Mycobacterium vaccine on the expression of Eotaxin mRNA in lung tissues of asthmatic guinea pig. Methods 30 guinea pigs were averagely divided into NS group, asthmu group and Mycobactefium vaccine group at random. A guinea pig model of asthma was formed with ovalbumin (OVA). Every guinea pig of Mycobacterium vaccine group was injected intramuscularly with 22.5 μg Mycobacterium vaccine 10 days before OVA immunization. After the last challenge, the numbers of inflammation cells in bronchoalveolar lavage fluids (BALF) were counted, pathological character of lung tissues was observed, the expression of Eotaxin mRNA in lung tissues was determined by in situ hybridization. Results The number of eosinophils in BALF, asthmatic airway inflammation response and expression of Eotaxin mRNA in lung tissues in Mycobacterium vaccine group were significantly lower than those of asthma group ( all P 〈 0.01 ). Conclusions Mycobacterium vaccine can decrease the number of eosinophils in BALF and lung tissues, which can inhibit the expression of Eotaxin mRNA in lung tissues of asthmatic guinea pig.
