摘要 目的 :用氧化低密度脂蛋白干预培养的人脐静脉内皮细胞 ,观察内皮细胞表达基质金属蛋白酶 - 2(MMP - 2 )的改变。方法 :用胶原酶消化法培养人脐静脉内皮细胞 ,分组后分别予以 2 5mg/LLDL、2 5mg/LOX -LDL进行干预。提取细胞总RNA和总蛋白分别进行RT -PCR、Westernblotting检测 ,同时收集条件培养基行酶谱法 (zy mography)检测MMP - 2的活性。结果 :经mRNA、蛋白和酶活性检测 ,发现 2 5mg/LOX -LDL干预时 ,MMP - 2的表达水平明显高于对照和 2 5mg/LLDL。结论 :OX -LDL能促进内皮细胞表达MMP - 2 ,这提示血管细胞外基质的降解在氧化脂蛋白诱发的动脉粥样硬化斑块产生和破裂机制中起着一定作用。 AIM: To investigate the effects of oxidized low density lipoprotein (OX-LDL) on expression of matrix metalloproteinase-2 (MMP-2) in human umbilical vein endothelial cells (HUVEC). METHODS: The MMP-2 mRNA was determined by reverse transcriptase-polymerase chain reaction (RT-PCR), MMP-2 protein was measured by western blotting and MMP-2 activity in conditioned medium was observed by gelatin zymography. RESULTS: In comparison with the control, there was no difference in the expression of MMP-2 between control and 25 mg/L LDL, whereas 25 mg/L OX-LDL increased MMP-2 activity in HUVEC ( P <0.01). CONCLUSION: OX-LDL enhances the expression and activity of MMP-2 in HUVEC, which may provide a explanation for the phenomenon that OX-LDL induces atherosclerotic lesions and plaque rapture.
