黄芪对梗阻性黄疸大鼠心肌TNFα及SOD基因mRNA表达的影响 被引量:10

摘要 目的 :探讨黄芪对梗黄心肌组织中TNFα、SOD基因mRNA表达的影响及分子生物学作用机制。 方法 :RT -PCR法扩增cDNA ,电泳扫描后半定量分析TNFα、SOD基因mRNA表达 ,组间比较。 结果 :黄芪可明显降低梗黄大鼠心肌组织中TNFα、mRNA基因表达 ,上调SODmRNA表达 ,降低血浆中TNFα水平 ,并使SOD合成增多 ,对梗黄心肌损害起到保护作用。 结论 :黄芪通过下调TNFαmRNA基因表达、上调SODmRNA表达、降低血清中TNFα水平 ,并使SOD合成增多 ,在多层次、多靶点通过双相调节作用 ,保护梗黄对心肌的损害。 Objective To detect the effect and significance of astragalus membranaceus(AM)on expression of TNFα and SOD gene mRNA in the injury of rat myocrdium with onstructive jaundice(OJ) Methods Semi-quantative RT-PCR analysis of TNFα and SOD mRNA expression in rat myocardium of OJ.Results When OJ occurred,the level of serum TNFα increased with the duration of the obstruction,but the expression of TNFα mRNA did not increase quickly,but obviously in OJ 12 days,the level of SOD in the myocardium decreased obviously during OJ.Expression of SOD mRNA was downregulated and decreased more obvioisly with the time passing on.AM could upregulate the expression of SOD mRNA,downregulate the expression of TNFα mRNA,so it could increase the synthesis of SOD in the myocardium and decrease the serum TNFα level.Conclusion Expression and synthesis of TNFα increased during OJ,and the time of synthesis was later than that of the increased level in serum.Expression of SOD mRNA was downregulated during OJ and the level in tissue decreased because of consumption.TNFα might participate in the course of injury of the organ in OJ through downregulating the expression of SOD.AM can protect the organ from OJ damage by downregulating the expression of TNFα mRNA and serum level,upregulating the expression and synthesis of SOD mRNA in multiple phases and targets from bi-directional regulation.
