摘要
目的 选择测定血清胆红素的最佳方法。方法 用重氮酸盐法、钒酸盐氧化法、胆红素氧化酶法对血清总胆红素 (TBI)和直接胆红素 (DBI)分别进行测定并比较。结果 3种方法重复性试验批内变异和批间变异分别小于 3 .0 %和 5 .0 % ;回收率均小于 5 .0 % ;重氮法和钒酸盐法测定TBI的线性范围为 0~ 70 0 μmol/L ,氧化酶法为 0~550 μmol/L ;3种方法测定DBI的线性范围为 0~ 40 0 μmol/L ;3种方法测定TBI、DBI,每 2种方法之间具有高度相关性。结论 钒酸盐法与氧化酶法、重氮酸盐法相比 ,测定TBI和DBI具有检测范围大 ,抗干扰能力强等特点 。
Objective To select the best method that detects bilirubin in serum.Methods The total bilirubin and direct bilirubin in serum were analyzed and compared by the methods of diazotate, vanadate oxidation and oxidase respectively.Result The aberrance in groups and that between groups of repetitive experiments of 3 kinds of methods were less than 3.0% and 5.0% respectively. The rates of recoveries were all less than 5.0%. The linear range of TBI measured by the methods of diazotate and vanadate oxidation was 0~700μmol/L, and that by the method of oxidase was 0~550μmol/L. The linear range of 3 kinds of methods to measure DBI was 0~400μmol/L. There was a strong relativity between either two methods to measure TBI and DBI. The anti interfernce ability of vanadate oxidation and oxidase was very strong. However, the method of diazotate was easily obstructed by some interferential factors.Conclusion The method of vanadate oxidation is more suitable in clinical normal check than oxidase and diazotate, because of the characteristics of the wide range of measurment and the anti interference ability.
