摘要 目的 建立用胆红素氧化酶 (BOD)作为催化剂 ,选择性地测定血清结合胆红素 (CB)的新方法。方法 测定条件为 :0 1mol/L甘氨酸缓冲液、pH 10 0 ,BOD活性 0 5~ 0 8U/反应管 ,反应时间 2分钟。结果 线性至少可达到 2 2 0 μmol/L ;精密度为批内CV 1 71~ 3 98%、批间CV 8 0~ 9 72 % ;正常参考值范围为 0~ 3 14μmol/L ;与偶氮法所测结果相关性较好。结论 本文所建立的CB测定方法具有良好的分析特异性 。 Objective A new enzymatic assay for selectively measuring conjugated bilirubin concentration in serum with use of bilirubin oxidase(BOD)was developed.Methods Conditions of the assay:0 1 mol/L glycine buffer,pH 10 0;BOD activity,0 5~0 8 U/tube;reaction time 2 min.Results Analytical index of this method:the linearity was extended to at least 220 μmol/L;the intra and inter assay CV s were 1 71%~3 98% and 8 0%~9 72% respectively;the reference values from 30 healthy individuals were 0~3 14 μmol/L;and the result of CB assay correlated well with that of DB assay based on the diazo method.Conclusion The proposed method for CB has good analytical specificity and is a reliable indicator of hepatobiliary excretion.
