摘要
目的 观察、评价白细胞介素 12 (IL - 12 )对结核分支杆菌感染小鼠的治疗结果和三种细胞因子水平的变化。方法 将BALB/C小鼠 5 0只 ,制成小鼠结核分支杆菌感染模型 ,随机分为对照组(PBS)和IL - 12共两组 (每组 2 5只 )。给予PBS或IL - 12治疗 ,ELISA法检测血清IFN -γ、IL - 4、IL- 10水平和器官菌落计数。结果 PBS组与IL - 12组的器官菌落计数 ,IFN -γ和IL - 10比较有显著性差异 (P <0 .0 1) ,而血清IL - 4水平变化不大 ,两组比较差异无显著性。结论 IL - 12诱导IFN -γ产生 ,促进Th1细胞分化成熟 ,改变了 /Th2平衡 。
Objective To study the effect of IL-12 on cytokines of mice infected with Mycobacterium tuberculosis. Methods 50 BALB/C mice infected with Mycobacterium tuberculosis were randomly divided into normal control (PBS) and IL-12 groups ,treated with PBS or IL-12 respectively .Blood level of IFN-γ IL-4 IL-10 was detected with ELISA method. The numbers of viable bacteria in organs were counted. Results There were significant differences between the two groups on INF IFN-γ IL-4 IL-10 ,and no change of the level of IL-4 in two groups.Conclusions IL-12 can induce IFN-γto be produced , to promote the development of Th1 cell responses, and to modulate the cytokine balance between Th1 and Th2, these suggest that IL-12 enhance the host's defense against the infection.
