摘要
目的 制备一种高活力的固定化胰蛋白酶 ,通过优化其制备条件提高活力回收。方法 将壳聚糖敷涂在硅胶的表面制成载体 ,以戊二醛为交联剂 ,通过交联反应将胰蛋白酶固定至载体上制得。对固定化后的滤液做回收再利用实验。结果 优化后制得的固定化胰蛋白酶的活力单位 (AU)高达 6 84 8.4U·g-1 ,活力回收 (AR)为 2 1 .8%。回收再利用实验得到的固定化胰蛋白酶活力单位仍高达 5 90 7.2 0U·g-1 。结论 制备的固定化胰蛋白酶活力单位高 ,活力回收也较高 。
OBJECTIVE To prepare an immobilized trypsin with high activity. After optimizing the preparing conditions, a higher activity retention was expected to be reached. METHODS Chitosan covered with silica as support and glutataldehyde as crosslinking agent were used to prepare immobilized trypsin. Re-utilization experiments were done with the liquid after immobilizing reaction. RESULTS After optimization the activity of immobilized trypsin reached 6848.40 U·g -1 ,and its activity recovery reached 21.8%. In the re-utilization experiments the activity of immobilized trypsin reached 5907.20 U·g -1 .CONCLUSION The activity units and activity recovery prepared immobilized trypsin were high, which is good for its application to industry.
