摘要
目的 从 TGF- β1 m RNA基因调控变化的角度阐明硒 (Se)和维生素 E(VE)缺乏对大鼠心脑组织损伤的作用机制。方法 通过 Se和VE缺乏的半合成人工饲料喂养大鼠 ,建立 Se和 VE缺乏疾病大鼠动物模型。并采用 RT- PCR检测 TGF-β1 在心脑组织中 m RNA表达变化特点。结果 低 Se低 VE组大鼠心肌可见散在微小灶状心肌坏死 ,脑组织海马、皮层可见神经细胞数量相对减少 ,部分神经细胞周围间隙增宽。 Se和 VE单项缺乏及联合缺乏 TGF-β1 m RNA表达均增高。以 Se和 VE联合缺乏变化最显著。结论 Se和 VE缺乏可诱导大鼠心脑组织 TGF-β1 m RNA表达上调 ,是缺血缺氧性心肌坏死和脑组织损伤的原因之一。 Se和 VE作为抗氧化微量营养素具有细胞保护作用 ,有实际应用价值。
Objective In order to discuss the effect of seleium (Se) and vitamin E (VE) deficiency on the mechanism of injury of rat heart and brain cells, especially to confirm the protecting cell and clinical value of Se and VE as antioxidant trace nutritional elements one point of view of alteration of TGFβ1 mRNA gene.Methods To establish the rat model of Se and VE deficiencies by feeding with mixture forage of Se and VE deficiency, and determine the expression of TGF-β1 in heart and brain tissues by RT-PCR method.Results The section of Se and VE deficiency group could be observed scattered focal necrosis of heart and brain tissues, decreased number of nerve cells has seen in hippocampus, cortical areas of brain, the width of circumstance space of nerve cells increased.Conclusions Deficiency of Se and VE and increase the expression of TGF-β1 mRNA, and it may be one of reasons of anoxemic myocardium necrosis and brain tissues lesion related with peripheral characteristic. Se and VE and protect heart and brain cells.
