A New Desiging Approach to Electrochemical Immuno-sensor Based on Electrostatic Reject of Gold Nanoparticles Modified with Alginates

摘要 通过胱胺将负电性的褐藻酸钠(AS)共价结合到纳米金表面形成聚合体,并以此来标记三碘甲状腺氨酸(T3).T3抗体固定在金电极上构成检测T3的电化学免疫传感器.通过免疫反应,标记T3和待检测的T3结合到金电极表面,使电极表面微环境发生显著改变,用Fe(CN)64-为电化学探针,用循环伏安法获取电极表面微环境改变的电化学信息来检测T3.其检测的线性范围为100~10 000 pg/mL,检测限为45 pg/mL. The aggregate, which was composed of gold nanoparticles modified with alginate via cystamine, was used as a label for T3. The electrochemical immuno-sensor was obtained by covalently immobilized antibody against T3 on the surface of gold electrode. Both labeled and analyte T3 were anchored on the immuno-sensor because of reaction between T3 and its antibody. This led to the changes of micro- environment of the surface of immuno-sensor. The changes, that were indicated by cyclic voltammetry technology using Fe (CN)64- as the electrochemical probe, was corresponding to the reaction between T3 and its antibody. The proposed immuno-sensor was successfully applied to monitor the concentration of T3. The dynamic range of determination was 100~10 000 pg/mL with a detection limit of defermination was 45 pg/mL.
