摘要
利用自组装膜技术在金电极表面形成3-巯基丙酸单分子膜,通过偶联剂将丙肝抗体与硫堇通过共价键键合在修饰电极上构建丙肝电化学免疫传感器,研究了传感器的响应特性,对阳性对照血清的检测,硫堇的峰电流有显著的下降,以硫堇的还原峰电流的下降百分数(k)作为检测丙肝抗原的依据。结果表明,k值与被测抗原在3.2~16μg/mL范围内成良好的线性关系,r=0.9951。将传感器应用于实际血清样品的检测,与ELISA检验结果相一致。
A novel HCV electrochemical immunosensor was fabricated by assembling 3-mercaptopropionic acid(3-MPA) on the gold electrode surface to bind the thionine and the HCV antibody. The CV vohammogram of the immunosensor displayed a couple of redox peaks of thionine. It was shown that the peak current decreased after HCV antigen reacting with the HCV antibody. The decrease percentage (k) of the reduction peak current was proportional to the concentration of the HCV antigen ranging from 3.2μg/mL to 16 μg/mL with a correlation coefficient of 0. 995 1. This immunosensor can differentiate the positive/negative standard human serum. The results obtained with the immunosensor for detecting HCV in human serum were in good agreement with those obtained by ELISA.
