以薄叶系文心兰Oncidiumaloha'Iwanaga'茎尖诱导形成的原球茎(PLB)为外植体,采用树脂膜培养容器(CP)的液体静置培养方式,比较不同碳源(蔗糖、麦芽糖、山梨醇)和不同有机添加物(蛋白胨、胰蛋白胨、苹果汁、番茄汁、椰子汁)对文心兰PLB增殖的影响.结果表明,以蔗糖为碳源时,PLB在鲜重和干物重方面显著高于麦芽糖和山梨醇处理,同时PLB分化幼苗数较少;不同有机添加物处理,蛋白胨对PLB增殖效果显著,其次分别为蕃茄汁>椰子汁>苹果汁>蛋白胨.在幼苗分化方面,胰蛋白胨、苹果汁和椰子汁处理中,幼苗分化数显著高于蛋白胨和番茄汁处理,并在处理间存在显著性差异.蛋白胨和番茄汁两处理间在鲜重、干物重和幼苗分化数方面无显著性差异.
By using protocormlike body (PLB.) induced from stalk tip of Oncidium Aloha 'Iwanaga' as material, and liquid still culture in film culture vessels as the culture mode, experiments were conducted to compare the effect of different carbon sources (Sucrose, Maltose and Sorbitol) and different organic compounds (Peptone,Tryptone,Coconut juice,Tomato juice and Apple juice) on the multiplication of protocormlike body (PLB).The results showed,when sucrose was used as carbon source, the fresh weight and dry matter weight of the PLB. after multiplication were evidently higher than when maltose and sorbitol were used as carbon sources, meanwhile differentiated shoots from the PLB were less. In terms of the effect of different organic compounds tredtment, peptone had the most significant effect on the PLB multiplication, next came tomato juice,coconut juice,apple juice and tryptone. As for the differentiation of shoots, the number of shoots in Tryptone, apple juice and coconut juice treatments were significantly higher than those in peptone and tomato juice treatments, and there existed significant difference between treatments. There existed no significant difference between peptone and tomato juice treatments in fresh weight, dry matter weight and the number of differentiated shoots.
