摘要
目的 观察尼莫地平(NIM)对5-羟色胺(5-HT)诱发的耳蜗微循环障碍的作用。方法 22只豚鼠随机分为A、B两组,A组于圆窗膜置放0.5%的5-HT,B组先于颈静脉输入0.5μg·kg-1·min-1剂量的NIM15 min,再于圆窗膜处置放0.5%的5-HT,同时继续输入该剂量的NIM。以激光多普勒流量仪测量耳蜗血流量(CBF)的变化。以压力传感器监测平均颈动脉血压(MABP)的变化。CBF信号和MABP信号同步输入计算机进行记录,供分析和统计学处理。结果 A组5-HT的运用使CBF出现了瞬时迅速下降和持续平稳下降两个时期的变化。B组NIM的运用使5-HT导致的耳蜗血流量的两个时期的下降分别得到了67.4%的抑制和完全的抑制。结论 NIM能有效地改善5-HT导致的耳蜗微循环障碍,增加耳蜗血流量。
Objective To investigate the effect of nimodipine (NIM) on ischemia cochlear induced by senotonin (5-HT). Methods Twenty-two guinea pigs were divided randomly into two groups: 12 in group A and 10 in group B. 2 μl 0.5% 5-HT were applied to round window membrance in group A, and 0. 5 μg·kg-1 · min -1 NIM was administed for 15 min through the cervical vein before the application of 5-HT on round window membrance and lasted throughout the experiment. The cochlear blood flow (CBF) was measured with laser doppler flowmeter. The mean arterial blood pressure (MABP) was recorded simultaneously. Results 5-HT induced a biphasic response of reduction in CBF in group A. In the first phase, CBF showed a transient abrupt reduction. The second phase reduced slowly. In group B, the transient reduction in CBF caused by the administration of NIM reached 67.4% and it inhibited the reduction in the second phase completely. Conclusion NIM can enhance the blood supply of ischemic cochlear induced by 5-HT.
