摘要
采用RT_PCR方法对分离自河北省某鸡场的一株新城疫病毒 (HBG_1)的核苷酸进行扩增 ,获得了分离株F0 裂解位点附近的基因片段 ,经测序分析 ,并与国际上已发表的新城疫病毒的核酸序列进行比较 ,结果表明其与标准株和疫苗株之间的同源性较低 ,仅为 82 %~ 86 %之间。运用计算机软件对其裂解位点处的氨基酸顺序进行预测和分析 。
In order to find the difference of the cleavage site of a strain with F 48 E 9 and C 30 ,a part of the cleavage site of Fusion protein gene fragment was amplified by RT_PCR.The sequence analysis showed this strain had low homology with F 48 E 9 and C 30 .Their homology were between 82 %~86 % in nucleotide sequence.A phylogenetic tree based on the published sequences of NDV reference strains was constructed and showed the isolated strain HBG_1 belonged to the genotype Ⅶ NDV,a novel genotype NDV.The amino acid sequence of the cleavage site region(residues 112~117) of the F protein of strain HBG_1 was matched to that of all virulent NDV strains,and the strain was proved to be an virulent strain.
