摘要
目的考察制备平均分子质量在 2 0 0 0~ 80 0 0间的低分子肝素钙的优化条件。方法以市售注射用肝素钠为原料 ,经过亚硝酸钠降解成低分子肝素钠 ,树脂脱钠 ,氧化钙中和转型成钙盐 ,最后用乙醇沉淀析出。结果最佳条件 :降解的pH值为 2 8,亚硝酸钠的浓度为 0 2 85mol/L ,氧化钙中和反应后 ,氧化钙加入的浓度为 8 0mol/L。结论该条件下制备的低分子肝素钙符合质量标准 。
Objective To investigate the optimum conditions of preparation of minihep calcium of which its molecular weights were between 3 000 and 8 000 . Methods Heparin of injection level was treated through degradation by sodium nitrite,desalting by 732 ion exchange resin, neutralization with calcium oxide, separation with alcohol precipitation. Result Optimum conditions: pH was 2.8,degradative concentration of sodium nitrite was 0 285 mol/L , the calcium chloride was 8 0 mol/L . Conclusion The minihep calcium prepared under the optimum conditions is coincidence with the standard sample, this indicates that this method of preparation is valuable.
