乙双吗啉合并用药的实验研究

摘要 乙双吗啉系我国自行合成的抗癌新药,它有抗肿瘤、抗转移、放疗增效及对抗阿霉素心脏毒性等方面的作用,细胞动力学研究证明,它可延长T_c、T_(G2)、T_3、使肿瘤细胞同步化于G_2期,这为合并用药提供了细胞动力学基础,本实验发现乙双吗啉可增强环磷酰胺、消瘤芥、AT-290、阿霉毒、争光霉素、氮烯咪胺及长春新碱等对S_(37)的抗肿瘤作用,与5-氟尿嘧啶、冬凌草甲素等合用呈相加效果,在非抗肿瘤药物中,乙双吗啉与依地酸钠合用呈协同效果。 Bimolane(AT-1727)is one of the bis-dioxopiperazine compunds. It has broad-sepetrum antitumor activity with higher chemotherapeutic index, antimetastatic and radiopotentiating activities. Bimolane also can prevent DOX-induced cardiotoxicity. Cytokinetically, bimolane can prolong times of life cycle,S and G_2 phases(Tc,Ts and TG_2)and partially synchronize tumor cells in G_2 phase resulting in the cytokinetic basis for combination chemotherapy. In this experiment,we found that bimolane could potentiate the antineoplastic action of some alkylating agents such as cyclophosphamide, AT-1258 and AT-290. The antitumor action of bleomycin,adriamycin,DTIC and vincristinc was synergized by bimolane. Additive antitumor effect was shown in the combination of bimolane with 5-FU,daunomycin and oridonin. The synergistic effect was found in the combination of bimolane with EDTA. The mechanisms of synergistic action of bimolane was discussed.
