摘要
在M9培养基的基础上,优选出重组大肠杆菌生产人干扰素-γ(rhIFN-γ)的优化培养基配方。结果表明:优化的碳源为含量2%的葡萄糖,氮源为蛋白胨和少量的(NH4)2SO4;优化培养基有利于菌体的生长和目标蛋白的表达,使rhIFN-γ的生产能力达0.64g/L,比优化前提高了190.9%。
Based on the selections of carbon and nitrogen contents,optimized prescription of culture medium was investigated for the fermentation of E.coli to produce recombinant human interferonγ.The optimum culture medium consisting of 2% glucose,tryptone and (NH4)2SO4 was found to benefit for the growth and expression of rhIFNγ,and to make the production level attain at 0.64 g/L which was enhanced by 190.9% in comparison with previous production level.
