摘要 目的:探索多份脐血混合移植重建免疫造血系统的可能性,扩大脐血移植的应用范围。方法:将1×10^6个C57BL/6和1×10^6个615小鼠脐血单个核细胞(MNC)同时输注给经致死量照射的BALB/c小鼠,10--15天后检查受鼠的脾结节(CFU—S),作受鼠的骨髓粒—巨噬细胞集落形成单位(CFU—GM)培养,以检测造血重建;1个月后用流式细胞仪(FCM)检测受鼠体内供者细胞的嵌合状态;用单向混合淋巴细胞反应(MLR)检测免疫重建和免疫耐受;用受鼠小肠和皮肤病理切片检测移植物抗宿主病(GVHD)。结果:混合脐血移植小鼠的生存率为65%,未移植小鼠全部死亡;移植后10一15天在小鼠的脾脏可以检测到CFU—S,小鼠骨髓培养有CFU—GM生长;移植后30天发现在同一个小鼠的体内有2个供者的不全嵌合体同时存在;受鼠对两个供鼠的淋巴细胞均产生了免疫耐受;受鼠小肠和皮肤病理切片检查表明有轻—中度GVHD。结论:混合脐血移植可以重建同种异基因小鼠的免疫和造血功能而不引起重度GVHD。本研究为扩大脐血移植的应用范围打下了一定的理论和实践基础。 Objective:To investigate the potential possibility to reconstitute the immunohematopietic function in mice by mixed near-term cord blood transplantation(CBT) , so as to lay a foundation for the clinical application of mixed CBT. Method: 1 ×106 fetal blood cells from 20-day-old C 57 BL/6 pups and 1 × 106 fetal blood cells from 20-day-old 615 pups delivered by cesarean were injected into a lethally irradiated BALB/c adult mouse in the same time. After 10-15 days, CFU-S and CFU-GM were examined to see whether hematopoiesis had been reconstituted. After one month. anti-H-2 Db and anti-H-2Kk McAb were used to investigate two donors' chimera in the recipients. In vitro mixed lymphocyte response (MLR) was used to investigate specific immune tolerance and immune reconstitution. The Pathological sections of intestine and skin from recipients were observed to investigate the gravity of graft versus host disease(GVHD). Result: All of the control mice died within three weeks after receiving lethal radiation. 65% recipients survived without serious GVHD within the observation period of two months. CFU-S and CFU-GM were found in the recipients on the 10-15th day after transplantation. Two donors ' mixed chimera was demonstrated in the same recipient. Immune tolerance and immune reconstitution were confirmed by in vitro MLR. Conclusion:Mixed CBT might reconstitute the immunohematopietic function in adult mice without causing serious GVHD. This model may be suitable to address many questions raised in clinical mixed CBT.
