摘要
以河豚毒素 (TTX)与中国鲎血蓝蛋白 (TTH)的化学偶联物TTX TTH免疫BALB/c小鼠。在免疫总剂量 (4 5 0 μg)相同时 ,比较长(L)、短 (S)不同免疫时间间隔 (分别为 14~2 0天和 7~10天 )对疫苗有效时间的影响 ,以血清抗体质量和抗TTX攻毒效果检验疫苗的效价。结果发现 ,S组比L组的抗体质量升速更快。L和S两组抗毒效价比较 :首次免疫后一年内 ,15次 (15×LD)攻毒 ,平均存活率分别为92 9%和 99 3% (P <0 0 1) ;半数动物存活时间分别为首次免疫后 11 3和 14 6个月。说明TTX的实验疫苗不同免疫方案均有效预防了TTX的攻毒 ,但短间隔比长间隔免疫产生更优的抗体质量 。
Tetrodotoxin(TTX) was coupled to Tachypleus tridentatus hemocyanin (TTH) chemically to form an artificial antigen.Balb/c mice were immunized with TTX TTH. The immunologic effect of different immunization schedules [14~20d for longer interval (group L) and 7~10d for shorter interval (group S)] of experimental anti TTX vaccine was compared. The quality of antisera and effect of vaccine on TTX poisoning were detected. The antibody quality of serum was increased more quickly in group S than that in group L after immunization. The average survival rates were 99 3% and 92 9%, respectively ( P <0 01)after 15 times of toxin challenge (15×LD) within one year of immunization, and 50% survival rate was abserved 11 3 months and 14 6 months, respectively, after immunization. The schedule S showed higher immune response than that of the schedule L. The experimental vaccine of TTX could effectively protect animal from TTX intoxication. The schedule S was better both in antibody quality and in the effect of TTX detoxification than that of the in schedule L. Therefore a shorter interval between immunization was more favorable to obtain a higher immunization affect in the case of TTX vaccine
