摘要
目的 构建霍乱弧菌抗菌抗毒口服活疫苗候选株 ,并在家兔模型中评价它们的免疫原性及主动保护力。方法 应用染色体 质粒致死平衡系统 ,在无毒的霍乱弧菌中表达霍乱毒素基因 ,继而在家兔模型中进行免疫原性的比较。结果 构建的分别含有突变无毒的ctx 和ct B基因的IEM10 4和IEM10 5菌株免疫家兔后 ,刺激机体产生抗菌与抗毒抗体 ,前者的特异性抗体滴度较后者高 ,家兔主动保护试验中对纯霍乱毒素 (CT)攻击保护前者也高于后者。结论 构建的霍乱疫苗候选株IEM10 4和IEM10 5在家兔模型中均能产生较好的抗菌与抗毒免疫 ,IEM10 4对家兔的免疫原性和主动保护力略高于IEM10 5。
Objective Development of live oral vaccine candidates against Vibrio cholerae. Methods Mutant ctx gene (ctx *) and ct B gene were separately expressed in Vibrio cholerae with a chromosome plasmid balanced lethal gene expression system. The vaccine candidates against Vibrio cholerae containing ctx * or ct B gene were constructed (IEM104 and IEM105) and their immunogenicity were compared in rabbit model. Results The specific antibody titre of IEM104 were higher than that of IEM105 after vaccination. IEM104 could neutralize 5μg CT which was higher than that by IEM105 (2μg) in rabbit ligated ileal loop system. Conclusion IEM104 and IEM105 appeared safety and superiority of antitoxic immunity to IEM101 in animal model.
