摘要
目的 建立毛细管电泳定量检测血清中维生素K1和K2的方法。方法 利用固相萃取方法提纯血清中维生素K ,将 2 0mmol/L的四硼酸钠、10mmol/L的SDS加入到含有 80 %甲醇、10 %去离子水、10 %乙腈的溶液中配置成缓冲液 ,在 2 5 4nm波长条件下对样本进行分离。结果 利用这种方法对血清中维生素K1和K2进行了有效的分离 ,该方法的线性低于病理状态下血清中维生素K的浓度 ,回收率为 97%~ 10 3% ,日内精密度 (CVs)和日间精密度分别为7.16 %~ 8.0 3%和 10 .0 8%~ 11.3%。结论 毛细管电泳方法测定血清中维生素K1和K2 ,具有快速、灵敏、定量检测等优点 ,可能成为测定人类血清中维生素K状态的有效检测方法。
Obejective To build an effect method to quantitatively analyze vitamin K in serum . Method A procedure for serum determination utilizes a solid-phase extraction cartridge(SPE). The analytes are separated by capillary electrophoresis with the methanol buffer containing 20 mmol/L sodium tetraborate, 10 mmol/L sodium dodecyle sulfate .PDA detector is monitored at 254nm. Results The phylloquinone and menaquinone in serum are separated using the new method. The assay linearity is far below physiological concentrations .The within-day and between-day coefficients of variation are 7.16%~8.03% and 10.08%~11.3%, respectively. The recovery is 97%~103%. Conclusion The quantitative analytical method is rapid and sensitive which may become a useful tool for the assessment of vitamin K status in humans.
