毛细管电泳检测唾液酸的方法及初步应用

摘要 目的 利用毛细管电泳 (CapillaryElectrophoresis ,CE)分离效果好、稳定性强的特点。建立毛细管电泳测定血清中唾液酸 (SialicAcid ,SA)的方法 (CE SA法 ) ,并与经典的化学比色法 (CH SA法 )进行对比研究 ,以达到快速、准确测定血清SA的目的。方法 在 2ml试管中分别加入 0 0 5mol/L硫酸 1ml和 10 0 μl血清 ,充分混合 ,然后试管紧塞 ,于 80℃水浴箱中水浴 30min ,试管冷却后离心 5min ,吸 1ml上清液于自动进样管中上机进样。结果 CE SA法检测范围为 1 2 0~2 80mmol/L ,线性方程为y =-0 .6 48± 6 .394x ,相关系数 0 .9993 ,平均回收率是 10 1.2 3 %。 113名交警唾液酸 (CE SA法 )均值为 (1.76± 0 .2 0 )mmol/L[(5 49.30± 3 .73)mg/L]。CH SA法测定均值为 (1.75± 0 .2 4)mmol/L ,两方法无显著性差异 (P>0 .0 5 )。结论 毛细管电泳能提供一种很好的分离、检测唾液酸的方法 。 Objective With its high sensitivity and reliability, a high performance capillary electrophoresis (HPCE) method was established to detect the serum sialic acids(SA) in the 113 traffic policemen, in comparison with the classical chemical method. Methods A 1 ml aliquot of 0 05 mol/L sulphuric acid was added to 100 μl of serum in a 2 ml test tube and vortex mixed, The test tube was tightly closed and heated at 80℃ for 30 min to hydrolyse the sample. After cooling, a 1 ml aliquot of the resulting solution was injected into the chromatograph. Results Peak showed at 4.14 min, the method was linear within the range of 1.20~2.80 mmol/L. The linear regression equation for the concentration versus peak area was y=-0.648+6.394 x, with a correlative coefficient of 0.999 3. The average recovery was 101.23%. The mean levels of SA in the 113 traffic policemen obtained by using HPCE and common chemical method respectively were (1.76±0.20) mmol/L[(549.3±63.73)mg/L] and (1.75±0.24)mmol/L. The two methods were not statistically significant ( P >0.05). Conclusion This method seemed to be simple with good separation efficiency, providing a suitable method for the SA analysis.
