摘要
目的 研究二甲基亚砜 (DMSO)对EC970 6细胞端粒酶活性及生长的影响。方法 用 1.3 %DMSO处理食管癌EC970 6细胞 ,TRAP -ELISA法检测细胞端粒酶活性的改变 ,流式细胞仪分析细胞周期的改变。结果 DMSO处理后的EC970 6细胞 ,端粒酶活性明显下降 ,且细胞周期明显改变 ,G1期细胞比率显著增高 ,S期细胞数显著降低。结论 DMSO可抑制EC970 6细胞端粒酶活性 ,使EC970 6细胞生长停滞于G1期 ,并诱导细胞凋亡。
Objective To Study the effects of dimethyl sulfoxide on telomerase activity and growth of EC9706 cells. Methods EC9706 cells were treated with 20 ml/L dimethyl sulfoxide (DMSO). Telomerase activity was measured using the telomeric repeat amplification protocol assay and enzyme-linked immunoadsordent assay (TRAP-ELISA), and cell cycle phases were analyzed by flow cytometry(FCM). Results Telomerase activity of treated cells was significantly decreased compared with the control group. FCM data showed that the G1 phase ratio was increased significantly and the S phase ratio was decreased obviously. Conclusion DMSO can inhibit telomerase activity of esophageal carcinoma EC9706 cells and block cell cycle progression at G1 phase, inducing cells apoptosis. These suggest that DMSO can be used in therapy of esophageal carcinoma.
