摘要
优化了花生总DNA的提取条件。用 3 .50ml细胞提取液 ,2 .50ml饱和KC1溶液 ,0 .2 5倍样液体积的酚 /氯仿 /异戊醇 (2 1∶2 8∶1 )溶液 ,0 .30倍样液体积的氯仿 /异戊醇溶液 ,0 .60倍体积的异丙醇 ,可从 1 0g花生芽中提得 4 .0mg纯DNA。用该法提取总DNA的产率高 ,时间短 。
The conditions of extracting total DNA from peanut is optimized.4.0mg of purified DNA was obtained from 1.0g peanut sprout under the following conditions:3.50ml cell extraction solution,2.50ml saturated KCl solution,the rate between the phenol/chloroform/isoamyl alcohol solution,chloroform/isomayl solution or isopropanol and the sample solution being 0.25,0.30and 0.60,respectively.
