摘要
目的 :研究骨髓基质干细胞 (bonemarrowstromalcells,BMSCs)经诱导、扩增后与冻干脱矿骨基质 (freeze -drieddemineralizedbonematrix ,fdDBM)的相容性及体外附着规律 ,为进一步体内回植提供研究基础。方法 :抽取成年犬骨髓 ,用全骨髓培养法获取单核细胞 ,经条件培养液体外诱导、扩增后 ,以不同浓度接种于制备的fdDBM上 ,通过倒置相差显微镜、扫描电镜观察细胞与材料的附着情况并以MTT实验了解适宜的接种浓度。结果 :当接种密度为 1.0× 10 6/cm2 时 ,细胞与支架材料之间有最大附着量。结论 :BMSCs能在fdDBM上粘附并生长 ,相容性良好 ,是理想的骨组织工程支架材料。
Objective:To study the biocompatibility and in vitro adhesion between freeze-dried demineralized bone matrix (fdDBM) and bone marrow stromal cells (BMSCs) that has been induced and proliferated, thus to provide a basis for their in vivo implantation.Methods:BMSCs obtained from canine bone marrow aspiration were in vitro induced and proliferated, then inoculated onto prepared fdDBM by different concentration.Their adhesion situation were analyzed by reverse phase difference microscope, scanning electro microscope and MTT method.Results:BMSCs were well adhered to fdDBM, the most appropriate inoculating concentration between them was 1.0×10 6/cm 2.Conclusions:BMSCs can be adhere to fdDBM and proliferate,indicating fdDBM is a potentially ideal scaffold material for bone tissue engineering.
