随机引物PCR法在变形链球菌基因分型中的初步应用 被引量:5

摘要 目的 :研究利用随机引物PCR法来对口腔变形链球菌进行基因分型。方法 :采用两组针对变形链球菌的随机引物 ,于不同的反应体系中 ,对变形链球菌群 (血清型c、e、f、d、g)及 3个临床分离株进行PCR扩增 ,并对变形链球菌的电泳图谱进行分析 ,鉴定其基因型的多态性。结果 :运用两组随机引物均可鉴别不同血清型的变形链球菌 ,对同一血清型的链球菌还可鉴别其不同的基因型。结论 :两组随机引物均能对变形链球菌进行基因分型。随机引物PCR法简便、快速、分辨率较高 ,是鉴定变形链球菌基因型的有效方法。 Objective:To study the genetic diversity within mutans streptococci (serotype c?e?f?d?g and 3 clinical strains).Methods:DNA was prepared from 7 reference strains of mutans streptococci together with 3 random clinical species certified as mutans streptococci. All the strains were genotyped by arbitrarily primed PCR with two different primers.Results:The result suggested that different serotypes showed different genotype patterns and there were even differences within the same serotype.Conclusions:Arbitrarily primed PCR analysis was useful to identify and even classify mutans streptococci.
