摘要 环氧化酶(COX)有两种同功酶,COX鄄1和COX鄄2。COX鄄2是一种诱导酶,在炎症和肿瘤的形成和发展中起重要作用。COX鄄2选择性抑制剂除对结肠癌、腺瘤息肉有化学预防作用外,还对胰腺癌、胃癌、食管癌、舌癌、皮肤癌、乳腺癌、膀胱癌、肺癌、前列腺癌等多种癌症有效。其可能的作用机理有的与抑制COX鄄2有关,如抑制PGE2生成、使底物花生四烯酸积聚、抗肿瘤血管形成等;有的与抑制COX鄄2无关,如下调Bcl鄄2、阻断Akt、上调Par鄄4、上调p27KIP1、释放细胞色素C等。
