摘要
目的 观察纳洛酮对内毒素刺激的大鼠离体支气管平滑肌细胞内皮素 1和内皮素A受体基因表达的影响。方法 分别用大肠杆菌内毒素O1 1 1 :B4 、内毒素和纳洛酮联合刺激离体培养的大鼠气管平滑肌细胞 8h后 ,应用反转录 聚合酶链反应 (RT PCR)方法检测其内皮素 1和内皮素A受体mRNA基因表达水平。结果 大鼠离体支气管平滑肌细胞在内毒素的作用下内皮素A受体mRNA基因高表达 ,同时还有一定量的内皮素 1mRNA基因表达 ;纳洛酮组大鼠离体支气管平滑肌细胞内皮素 1和内皮素A受体基因表达均明显减少。结论 内毒素可刺激大鼠支气管平滑肌细胞高表达内皮素 1和内皮素A受体基因 ;纳洛酮对内毒素刺激的大鼠离体支气管平滑肌细胞内皮素
Objective To investigate the effect of naloxone on the expression of ET 1 and ET AR mRNA in cultured rat bronchial smooth muscle cells stimulated by LPS.Methods The reverse transcription-polymerase chain reaction(RT PCR)method was used to detect the expression of ET 1 and ET AR mRNA of the cultured rat bronchial smooth muscle cells.Cells were randomized into normal group,control group(LPS group),naloxone group,incubated for 8 hours in 5%CO 2.Results The expression of ET 1 and ET AR mRNA increased significantly in control group,with no expression in normal group,but decreased markedly in naloxone group.Conclusion ET AR mRNA and ET 1 mRNA may be located in the cultured rat bronchial smooth muscle cells.LPS may,at transcription and translation levels,lead to the increase in synthesis and release of ET 1 mRNA and ET AR mRNA.Naloxone may,at transcription and translation levels,extend opposite effects.
