摘要
目的 研究双氯芬酸钠滴眼液对人晶状体上皮细胞增殖的抑制作用。方法 分离培养人晶状体上皮细胞 ,在培养液内加入双氯芬酸钠滴眼液 ,并以高三尖杉酯碱作为对照。测定药物对细胞的半效抑制量 ;观察药物对细胞贴壁和形态学的影响。结果 双氯芬酸钠滴眼液作用 2 4h对晶状体上皮细胞半效量ID50 为 0 78μg/ml,72hID50 为0 63 μg/ml,作用时间对细胞的抑制率无明显差异 (P >0 0 5 ) ;与对照组相比 2 4h的ID50 无显著差异 (P >0 0 5 ) ,而 72hID50 有明显差异 (P <0 0 5 )。结论 双氯芬酸钠滴眼液对培养人晶状体上皮细胞在体外试验有抑制作用 。
ObjectiveTo study the effects of diclofenac sodium drops on the inhibition of human lens epithelial cells (HLEC) proliferation in order to find a medicine which could prevent PCO.MethodsHLEC were isolated,cultured,and then exposed to different concentrations of diclofenac sodium.Homoharringtonine was used as positive control.After 24 h and 72 h incubation, ID 50 of diclofenac sodium and homoharringtonine were measured,the morphological changes and the rate of attached cells were observed.ResultsIt was showed that the medicine induced the cells widen,empty bubbles of the cytoplasm,and the nucleus shrunk.Compared to homoharringtonine group,there was no difference in ID 50 at 24 h,but the difference was significant in 72 h.The result showed that diclofenac sodium inhibited the proliferation of HELC but the effect be less than homoharringtonine.ConclusionDiclofenac sodium may inhibit proliferation of HLEC in tissue culture.The diclofenac sodium drop may be an effective medicine for prevention of PCO.
