摘要
目的:体外试验研究三氧化二砷(As2O3)对兔晶体上皮细胞(RLECs)增殖的影响。方法:取第3代RLECs,分为正常组和加入不同浓度三氧化二砷的实验组,分别于作用24小时、72小时和5天后观察各组LECs的形态学改变,用MTT比色法测定As2O3对LECs的影响。结果:As2O3作用于LECs24小时的IC50为3.101μmol/L,72小时的IC50为1.743μmol/L,5天的IC50为1.094μmol/L。结论:As2O3为剂量依赖性和时间依赖性药物,对体外培养的LECs增殖有明显抑制作用。
Objective: To evaluate The effect of arsenic trioxide on the proliferation of rabbit lens epithelial cells in vitro. Methods: Divide 3-passage RLECs into joined control group and different concentrations of arsenic trioxide groups for 24 hours, 72 hours and 5 days, the effects of RLECs were measured with methyl thiazolyl tetrazolium(MTT) assay and the apoptosis of cells were observed. Results: The IC50 of arisenic trioxide exposed to RLECs for 24 hours was 3.101μmol/L, that of 72 hours was 1.743μmol/L and that of 5 days was 1.094μmol/L. Conclusion: Arsenic trioxide was dose-dependent and time-dependent inhibitor, and inhibited the proliferation of RLECs in vitro effectively.
