目的 :探讨p16 ,p2 1和CyclinD1mRNA在自体移植静脉的表达及其与平滑肌细胞 (SMC)增殖的关系 .方法 :以新西兰兔建立自体移植静脉SMC增殖模型 ,用增殖细胞核抗原 (PCNA)免疫组化染色观察SMC增殖水平 ,用原位杂交观察p16 ,p2 1和CyclinD1mRNA表达水平 .结果 :p16mRNA在正常颈静脉无表达 ,移植后仅少量表达 ,与内膜SMC增殖无相关性 .p2 1mRNA在正常颈静脉低水平表达 ,4~ 8wk表达明显增加 ,12wk降低 ,表达水平在 1~ 8wk与内膜SMC增殖呈负相关 (r =- 0 .90 8,P <0 .0 1) .CyclinD1mRNA 1~ 2wk为表达高峰 ,4wk后逐渐下降 ,表达水平与内膜SMC增殖呈正相关 (r =0 .95 1,P <0 .0 1) .结论 :p2 1和CyclinD1mRNA表达与自体移植静脉内膜SMC增殖有明显相关性 ,p2 1可能对自体移植静脉内膜SMC增殖起抑制作用 ,Cy
AIM: To observe the expression of cell cycle regulating genes p16, p21 and CyclinD 1 in autologous vein graft (AVG) and its relation with proliferation of smooth muscle cell (SMC). METHODS: A model of proliferation of SMC in AVG was established in New Zealand rabbits. Proliferating cell nuclear antigen (PCNA) on different time spots post vein graft implantation was detected with immunohistochemistry and the expression of p16, p21 and CyclinD 1 mRNA in neointima of AVG was observed. Their correlation with the proliferation of SMC was analyzed. RESULTS: No expression of p16 mRNA was observed in the normal veins. Lower level expression was seen 1~2 wk postoperation and there was no correlation between the expression level of p16 mRNA and the proliferation of SMC. Lower level expression of p21 mRNA was detected in the normal veins. The expression increased 4~8 wk post implantation and decreased significantly in 12 wk. The expression level of p21 mRNA in intima (1~8 wk) was correlated negatively with the proliferation of SMC. Lower level expression of CyclinD 1 was observed in the normal veins. The peak level of expression occurred in 1~2 weeks post implantation. The trend of this expression was correlated positively with the proliferation of SMC. CONCLUSION: The results suggest that p16 has no regulatory effects on the proliferation of SMC of vein grafts. p21 inhibits the proliferation, while CyclinD 1 stimulates this process.
